Characterization of the Gene for 47Kd Protein of Rickettsia tsutsugamushi

Abstract
The gene for 47Kd protein of Rickettsia tsutsugamushi strain Kato was cloned and sequenced to provide the basic information for the vaccine development of tsutsugamushi desease.
Genomic library of Rickettsia tsutsugamushi was constructed in lambda ZAPII and screened with mouse anti-Rickettsia tsutsugamushi Kato sera. The insert DNA from a positive clone was subcloned into the PBluescript plasmid. The recombinant plasmid,
pKT5,
was revealed to contain the complete gene for the 47Kd prtein of Rickettsia tsutsugamushi. An open reading frame of 1.4Kb and the promoter region were identified by DNA sequencing and primer extension assay. Shine-Dalgarno sequence and the
translation
initiation codon were also located. The amino acid sequence that was deduced from the nucleotide sequence of the gene was determined to have 466 amino acid residues with the predicted molecular weight of 50.8Kd. The putative mature protein
without
the
leader peptide composed of 32 amino acids had the molecular weight of 47.2Kd.
